"Earlier this year, when Germans were cracking open boxes of chocolate muesli—a common breakfast cereal in their country—it's unlikely their thoughts slid to the chemical 4-methylbenzophenone, much less to the fact that this component of printing ink had slipped from the outside of the cardboard box and into the cereal. That is, until the European Food Safety Authority (EFSA) was asked to look into the matter.

Open a cereal box or a carton of juice, breathe in an asthma drug from an inhaler, or pop an antihistamine pill out of a plastic pouch's metal foil. You are probably thinking about the product you're about to consume and not about its packaging—except, perhaps, for a twinge of regret about contributing to landfill waste. But here's something to keep in mind: Even when the wrapping comes off, you inevitably ingest some of the container."

Acute effects of sulfur mustard injury--Munich experiences.

Acute effects of sulfur mustard injury--Munich experiences.

Toxicology. 2009 Sep 1;263(1):3-8. Epub 2009 May 29.

Bundeswehr Institute of Pharmacology and Toxicology, Neuherbergstr. 11, D-80937 Munich, Germany. kai.kehe@lrz.uni-muenchen.de

Sulfur mustard (SM) is a strong vesicant agent which has been used in several military conflicts. Large stockpiles still exist to the present day. SM is believed to be a major threat to civilian populations because of the persistent asymmetric threat by non-state actors, such as terrorist groups, its easy synthesis and handling and the risk of theft from stockpiles. Following an asymptomatic interval of several hours, acute SM exposure produces subepidermal skin blisters, respiratory tract damage, eye lesions and bone marrow depression. Iranian victims of SM exposure during the Iran-Iraq (1984-1988) war were treated at intensive care units of 3 Munich hospitals. All 12 patients were injured following aerial attacks with SM filled bombs, which exploded in a distance between 5 and 30m. All patients soon noted an offensive smell of garlic, addle eggs or oil roasted vegetables. No individual protective equipment was used. Eye itching and skin blistering started 2h after SM exposure. Some patients complained of nausea, dizziness and hoarseness. 4h after exposure, most patients started vomiting. Eye symptoms worsened and most patients suffered from temporary blindness due to blepharospasm and lid oedema. Additionally, pulmonary symptoms such as productive cough occurred. Patients were transferred to Munich 4-17 days after SM exposure. On admission all patients showed significant skin blistering and pigmentation. Conjunctivitis and photophobia were the major eye symptoms. Pulmonary symptoms, including productive cough were persistent. Bronchoscopy revealed massive inflammation of the trachea with signs of necrosis. 3 patients needed tracheotomy. Chest X-ray did not yield abnormal observations. This presentation summarizes the experience of treating SM victims in Munich and discusses therapeutic implications.

The chronic effects of sulfur mustard exposure.

The chronic effects of sulfur mustard exposure.

Toxicology. 2009 Sep 1;263(1):9-11. Epub 2009 May 30.

Organization for the Prohibition of Chemical Weapons, Johan de Wittlaan 32, 2517 JR, The Hague, The Netherlands.

Whilst the acute effects of sulfur mustard have been relatively well characterised, the chronic effects of short term but significant exposures are still evolving. The approximately 30,000 Iranian victims of CW exposure from the 1980 to 1988 Iran-Iraq war who are currently being followed form a key population who are now 20 years post-exposure. The key chronic findings in this population reflect the common acute effects of sulfur mustard, and are related to the skin, eye and respiratory system. Excluding pruritis, skin changes appear to settle. Eye symptoms are slowly progressive, however a severe, rapid onset form of keratitis is seen to develop in a number of patients after a latent period of 15-20 years. The respiratory tract also shows progressive deterioration, with bronchiolitis obliterans now being considered the main pathological feature of "mustard lung". In addition, there are other potential effects of sulfur mustard exposure which become evident only in the longer term and which are being investigated, including the development of cancer, immunological and neuropsychiatric changes, and reproductive effects. Finally, a chronic effect of sulfur mustard exposure that is now becoming apparent is the wider long-term social and economic effects of these illnesses on individuals and their families.

Received 7 November 2008;

Available online 5 March 2009.

Abstract

During recent years there has been increasing public concern on potential cancer risks from microwave emissions from wireless phones. We evaluated the scientific evidence for long-term mobile phone use and the association with certain tumors in case–control studies, mostly from the Hardell group in Sweden and the Interphone study group. Regarding brain tumors the meta-analysis yielded for glioma odds ratio (OR) = 1.0, 95% confidence interval (CI) = 0.9–1.1. OR increased to 1.3, 95% CI = 1.1–1.6 with 10 year latency period, with highest risk for ipsilateral exposure (same side as the tumor localisation), OR = 1.9, 95% CI = 1.4–2.4, lower for contralateral exposure (opposite side) OR = 1.2, 95% CI = 0.9–1.7. Regarding acoustic neuroma OR = 1.0, 95% CI = 0.8–1.1 was calculated increasing to OR = 1.3, 95% CI = 0.97–1.9 with 10 year latency period. For ipsilateral exposure OR = 1.6, 95% CI = 1.1–2.4, and for contralateral exposure OR = 1.2, 95% CI = 0.8–1.9 were found. Regarding meningioma no consistent pattern of an increased risk was found. Concerning age, highest risk was found in the age group <20 years at time of first use of wireless phones in the studies from the Hardell group. For salivary gland tumors, non-Hodgkin lymphoma and testicular cancer no consistent pattern of an association with use of wireless phones was found. One study on uveal melanoma yielded for probable/certain mobile phone use OR = 4.2, 95% CI = 1.2–14.5. One study on intratemporal facial nerve tumor was not possible to evaluate due to methodological shortcomings. In summary our review yielded a consistent pattern of an increased risk for glioma and acoustic neuroma after >10 year mobile phone use. We conclude that current standard for exposure to microwaves during mobile phone use is not safe for long-term exposure and needs to be revised.

Modulation of Cytokine Expression in Human Myeloid Dendritic Cells by Environmental Endocrine Disrupting Chemicals Involves Epigenetic Regulation

Modulation of Cytokine Expression in Human Myeloid Dendritic Cells by Environmental Endocrine Disrupting Chemicals Involves Epigenetic Regulation

Abstract:
Background: Exposure to environmental endocrine-disrupting chemicals (EDCs) is often
associated with dysregulated immune homeostasis, but the mechanisms of action remain unclear.

Objectives: The aim of this study was to test a hypothesis that EDCs regulate the functions of
human dendritic cells (DCs), a front-line, immunoregulatory cell type in contact with the
environment.

Methods: Circulating myeloid DCs (mDCs) from five subjects were investigated, and their
responses, with or without co-culture with autologous T cells, to two common EDCs, nonylphenol
(NP) and 4-octylphenol (4-OP), were measured. EDC-associated cytokine responses, signaling
events and histone modifications were examined by using ELISA, western blotting and chromatin
immunoprecipitation (ChIP) assay, respectively.

Results: In all cases, mDCs treated with NP or 4-OP demonstrated increased expression of TNF-α,
but decreased baseline and LPS-induced IL-10 production, which was partially reversible by an
estrogen receptor antagonist. Activation of the MKK3/6-p38 signaling pathway marked the effect
of NP on TNF-α expression, concomitant with enhanced levels of methyltranferase complex (MLL
and WDR5) in the nucleus, and of trimethylated H3K4, acetylated H3 as well as H4 at the TNFA
gene locus. Further, upregulated TNF-α expression was significantly suppressed in NP-treated
mDCs by a histone acetyltransferase inhibitor. In the presence of NP-treated mDCs, T cells showed
increased levels of IL-13, but decreased expression of IFN-γ.

Conclusions: These results suggest that NP and 4-OP may have functional effects on mDC's
response via, in part, the estrogen receptor, MKK3/6-p38 MAPK signaling pathway and histone
modifications, with subsequent influence on the T-cell cytokine responses.

of Cardiology and Division of Nuclear Medicine (Dr Ruddy, Ms Dalipaj),
University of Ottawa Heart Institute, Ottawa, Ontario, Canada.

OBJECTIVE:: To investigate the associations between exposure to particulate
air pollution and changes in cardiovascular function and plasma mediators in
seniors.

METHODS:: We monitored daily indoor and outdoor black carbon and particulate
matter </=2.5 mum (pm2.5) and personal pm2.5 repeatedly for 28 nonsmoking
seniors. We then measured their blood pressure, heart rate, and brachial
artery function, and determined plasma mediators of inflammation, vascular
function, and oxidative stress. We tested associations using mixed-effects
models.

RESULTS:: Increases in black carbon and pm2.5 were associated with increases
in blood pressure, heart rate, endothelin-1, vascular endothelial growth
factor, and oxidative stress marker thiobarbituric acid reactive substances,
and a decrease in brachial artery diameter (P < 0.05).

CONCLUSION:: Daily exposure to particulate pollution, likely
traffic-related, may result in adverse effects on cardiovascular function
and blood mediators that modulate vascular system in seniors.

Alpha-lipoic acid (LA), the metabolic antioxidant, was evaluated for its potential to protect against Cr(6+)-induced DNA damage. Potassium dichromate was administered to Swiss albino mice orally ad libitum at the doses of 5, 10 or 25 mg/kg body weight in drinking water to set DNA damage in cells, which was characterized in mouse peripheral blood mononuclear cells and bone marrow cells using single-cell gel electrophoresis and analyses of generated comets for Tail moment, Tail DNA and Tail length. DNA damage was dose dependent. Cytoprotection by LA was remarkable. LA (5, 10 and 25 mg/kg body weight intraperitoneally) in pre-, co- and post-toxicant administration schedule abrogated DNA damage substantially in both cell types. Protection by LA was also dose dependent. LA annulled DNA damage by Cr(6+) in plasmid relaxation assay. A negligible DNA damage resulted during interaction of Cr(6+) and LA. Compared to ascorbate, LA emerged as a better antioxidant and least DNA damaging. In conclusion, our study advocated an experimental therapeutic research potential in LA against Cr(6+)-induced DNA damage for reduction of occupational cancer risk in humans.

Debating How Much Weed Killer Is Safe in Your Water Glass
http://www.nytimes.com/2009/08/23/us/23water.html?_r=1&pagewanted=print

"For decades, farmers, lawn care workers and professional green thumbs have relied on the popular weed killer atrazine to protect their crops, golf courses and manicured lawns.  But atrazine often washes into water supplies and has become among the most common contaminants in American reservoirs and other sources of drinking water.  Now, new research suggests that atrazine may be dangerous at lower concentrations than previously thought. Recent studies suggest that, even at concentrations meeting current federal standards, the chemical may be associated with birth defects, low birth weights and menstrual problems."

A cohort study of the association between secondary sex ratio and parental exposure to polybrominated biphenyl (PBB) and polychlorinated biphenyl (PCB).

Environ Health. 2009 Aug 15;8(1):35. [Epub ahead of print]

ABSTRACT: BACKGROUND: Polybrominated biphenyl (PBB), a brominated flame retardant, was accidently mixed into animal feed in Michigan (1973-1974) resulting in human exposure through consumption of contaminated meat, milk and eggs. Beginning in 1976 individuals who consumed contaminated products were enrolled in the Michigan Long-Term PBB Study. This cohort presents a unique opportunity to study the association between parental exposures to PBB and offspring sex ratio. METHODS: We identified offspring of female PBB cohort participants (born 1975-1988) and obtained electronic birth records for those born in the state of Michigan. We linked this information to parental serum PBB and PCB concentrations collected at enrollment into the cohort. We modeled the odds of a male birth with generalized estimating equations accounting for the non-independence of siblings born to the same parents. We explored potential confounders: parental age and education at offspring's birth, parental body mass index at cohort enrollment, birth order, gestational age and year of offspring's birth. RESULTS: The overall proportion of male offspring among 865 live births to cohort mothers was 0.542. This was higher than the national male proportion of 0.514 (binomial test: p=0.10). When both parents were in the cohort (n=300), we found increased odds of a male birth with combined parents' enrollment PBB exposure [greater than or equal to] the median concentrations (3 ug/L for mothers; 6 ug/L for fathers) compared to combined parents' PBB exposure < the median concentrations (AOR=1.43, 95% CI: 0.89-2.29), although this did not reach statistical significance. In addition, there was a suggestion of increased odds of a male birth for combined parents' enrollment PCB exposure [greater than or equal to] the median concentrations (6 ug/L for mothers; 8 ug/L for fathers) compared to combined parents' enrollment PCB exposure < the median concentrations (AOR=1.53, 95% CI: 0.93-2.52). CONCLUSIONS: This study adds to the body of literature on secondary sex ratio and exposure to environmental contaminants. In this population, combined parental exposure to PBBs or PCBs increased the odds of a male birth. Further research is needed to corroborate these findings and shed light on the biological mechanisms by which these types of chemicals may influence the secondary sex ratio.

OBJECTIVE: To observe the effect of acupuncture on the fatigue degree in patients with chronic fatigue syndrome (CFS). METHODS: Seventy CFS patients were equally randomized into control and treatment groups according to randomized block design. Acupuncture was applied to Baihui (GV 20), Danzhong (CV 17), Zhongwan (CV 12), etc., for patients in treatment group, and to non-acupoints (2 cm respectively to the abovementioned acupoints) for those in control group. The treatment was given once every other day, 14 times altogether. The fatigue degree and the therapeutic effect were assessed by Chalder's fatigue scale (FS). RESULTS: A total of 64 cases (32/group) were finished in this study. After the treatment, the physical FS (5.0 +/- 2.4 vs 6.8 +/- 1.5), mental FS (1.8 +/-1.8 vs 3.1 +/- 1.5) and the total FS (6.8 +/- 3.8 vs 9.9 +/- 2.5) in treatment group, physical FS (5.0 +/- 2.5 vs 6.4 +/- 1.5) and the total FS (7.5 +/- 3.4 vs 9.6 +/- 2.8) in control group decreased significantly compared with pre-treatment (P < 0.01, P < 0.05). There was no marked change in mental FS (2.5 +/- 11.6 vs 3.2 +/- 11.6) in control group after the treatment (P > 0.05). Comparison between two groups showed no significant differences in the 3 indexes (P > 0.05). CONCLUSION: Acupuncture can relieve CFS patients' physical and mental fatigue and the therapeutic effect of acupuncture of acupoints is relatively better than that of non-acupoints in reducing mental fatigue.

Diabetic nephropathy is characterized by increased oxidative stress and
mitochondrial dysfunction. In the present study, we prepared primary
cultures of proximal tubular (PT) cells from diabetic rats 30 days after an
ip injection of streptozotocin and compared their susceptibility to oxidants
(tert-butyl hydroperoxide, methyl vinyl ketone) and a mitochondrial toxicant
(antimycin A) with that of PT cells isolated from age-matched control rats,
to test the hypothesis that PT cells from diabetic rats exhibit more
cellular and mitochondrial injury than those from control rats when exposed
to these toxicants. PT cells from diabetic rats exhibited higher basal
levels of reactive oxygen species (ROS) and higher mitochondrial membrane
potential, demonstrating that the PT cells maintain the diabetic phenotype
in primary culture. Incubation with either the oxidants or mitochondrial
toxicant resulted in greater necrotic and apoptotic cell death, greater
evidence of morphological damage, greater increases in ROS, and greater
decreases in mitochondrial membrane potential in PT cells from diabetic rats
than in those from control rats. Pretreatment with either the antioxidant
N-acetyl-L-cysteine or a catalase mimetic provided equivalent protection of
PT cells from both diabetic and control rats. Despite the greater
susceptibility to oxidative and mitochondrial injury, both cytoplasmic and
mitochondrial glutathione concentrations were markedly higher in PT cells
from diabetic rats, suggesting an upregulation of antioxidant processes in
diabetic kidney. These results support the hypothesis that primary cultures
of PT cells from diabetic rats are a valid model in which to study renal
cellular function in the diabetic state.

quetzl@rocketmail.com

Environmental illness (EI) still remains something of an enigma, despite attempts to squeeze it into the increasingly flexible framework of the biomedical model or label it as 'psychogenic'. Consistently, environmentally ill bodies fail to respond to conventional medical tests, with the emergence of ambiguous, indecipherable or negative results. During a series of in-depth interviews in Canada, both patients and environmental health practitioners advocated the need for an alternative paradigm from which to view EI. They also spoke of the complex nature of the illness, a conundrum that is currently being explored at the Nova Scotia Environmental Health Centre through an alternative epistemological framework: complexity theory. In this paper, I present a tentative proposal, based on the juxtaposition of theoretical insights from this new science to discourses emanating from patients and doctors. Specifically, I argue that the diverse symptoms of EI can be comprehended through a study of complexity theory, a conception that has positive implications for treatment.

Glyphosate-based herbicides are toxic and endocrine disruptors in human cell lines

Glyphosate-based herbicides are toxic and endocrine disruptors in human cell lines.
Gasnier C, Dumont C, Benachour N, Clair E, Chagnon MC, Séralini GE.
Toxicology. 2009 Aug 21;262(3):184-91. Epub 2009 Jun 17.

Glyphosate-based herbicides are the most widely used across the world; they are commercialized in different formulations. Their residues are frequent pollutants in the environment. In addition, these herbicides are spread on most eaten transgenic plants, modified to tolerate high levels of these compounds in their cells. Up to 400 ppm of their residues are accepted in some feed. We exposed human liver HepG2 cells, a well-known model to study xenobiotic toxicity, to four different formulations and to glyphosate, which is usually tested alone in chronic in vivo regulatory studies. We measured cytotoxicity with three assays (Alamar Blue, MTT, ToxiLight), plus genotoxicity (comet assay), anti-estrogenic (on ERalpha, ERbeta) and anti-androgenic effects (on AR) using gene reporter tests. We also checked androgen to estrogen conversion by aromatase activity and mRNA. All parameters were disrupted at sub-agricultural doses with all formulations within 24h. These effects were more dependent on the formulation than on the glyphosate concentration. First, we observed a human cell endocrine disruption from 0.5 ppm on the androgen receptor in MDA-MB453-kb2 cells for the most active formulation (R400), then from 2 ppm the transcriptional activities on both estrogen receptors were also inhibited on HepG2. Aromatase transcription and activity were disrupted from 10 ppm. Cytotoxic effects started at 10 ppm with Alamar Blue assay (the most sensitive), and DNA damages at 5 ppm. A real cell impact of glyphosate-based herbicides residues in food, feed or in the environment has thus to be considered, and their classifications as carcinogens/mutagens/reprotoxics is discussed.

The U.S. Department of Housing and Urban Development in collaboration with
the United States Environmental Protection Agency conducted a survey
measuring lead, allergens, and insecticides in a randomly selected
nationally representative sample of residential homes. Multistage sampling
with clustering was used to select the 1131 homes of which a subset of 500
randomly selected homes included the collection of hard surface floor wipes.
Samples were collected by trained field technicians between June 2005 and
March 2006 using isopropanol wetted wipes. Samples were analyzed for a suite
of 24 compounds which included insecticides in the organochlorine,
organophosphate, pyrethroid and phenylpyrazole classes, and the insecticide
synergist piperonyl butoxide. The most commonly detected were permethrin
(89%), chlorpyrifos (78%), chlordane (64%), piperonyl butoxide (52%),
cypermethrin (46%), and fipronil (40%). Mean and geometric mean (GM)
concentrations varied widely among compounds, but were highest for
trans-permethrin (mean 2.22 ng/cm2 and GM 0.14 ng/ cm2) and cypermethrin
(mean 2.9 ng/cm2 and GM 0.03 ng/ cm2). Results show that most floors in
occupied homes in the U.S. have measurable levels of insecticides that may
serve as sources of exposure to occupants.
